Most likely. With true free squatting (and even if using a smith machine), you are using most of your body to support the weight as you move up and down (shoulders, arms, back, abdomen, thighs, hamstrings, calves, etc.)

One way that I can think of to get huge quads like that without other parts of the body getting huge would be a leg extension machine:
